General Information: Dulbecco’s MEM (DMEM) Ham’s F-12 w/o HEPES, Glucose, AminoAcids, Linoleic Acid, Lipoic Acid, is the most widely used modification of BME. It containsa 4-fold higher concentration of amino acids and vitamins. Non-essential amino acids andcertain essential trace elements were added. The bicarbonate concentration was increased.The standard formula for DMEM is with 1000mg/ml glucose. DMEM “High Glucose“ with4500mg/ml aides in the cultivation of certain cell types. DMEM was originally developedfor the culture of mice embryonic cells. Today, it finds a broad application of serum freeculture of normal and transformed mouse and chicken cells.Ham’s Nutrient Mixtures were originally developed to support growth of several clones ofChinese hamster ovary (CHO) cells, of HeLa and of mouse L-cells. Ham’s mixtures wereformulated for use with or without serum supplementation, depending on the cell typebeing cultured. Ham’s F-12 was developed for growth of primary rat hepatocytes and ratprostate epithelial cells. It is also used in a clonal toxicity assay using CHO cells. Themedium is for research use only and not approved for human or animal use or forapplication in any in vitro diagnostic procedure. Storage of the medium andsupplements Please note that the basal medium has to be stored at 4°C up to 8°C butnever should be frozen. The supplement has to be stored at -20°C. Protect from light fora longer time period.Preparing and Stability:Thaw the supplement mixture at 37°C before adding to the basal medium. After thawingplease rinse the bottle and the tube with 70% ethanol as protection. Remove ethanolexcess before opening the cap of the tube. After adding the supplement mixture to thebasal medium slew carefully to get a homogenous medium. Please be careful not to touchthe tube or bottle interior threads with your fingers. Please avoid long term exposure tolight because some components are light sensitive. Also do not warm up the medium over37°C. All this protects your medium against contamination or degradation of essentialcomponents.
