General Information: Dulbecco’s MEM (DMEM) Ham’s F-12 w/o L-Alanine, L-Asparagine,L-Aspartic, L-Glutamic Acid, Glycine, L-Proline and L-Serine is the most widely usedmodification of BME. It contains a 4-fold higher concentration of amino acids andvitamins. Non-essential amino acids and certain essential trace elements were added. Thebicarbonate concentration was increased. The standard formula for DMEM is with1000mg/ml glucose. DMEM “High Glucose“ with 4500mg/ml aides in the cultivation ofcertain cell types. DMEM was originally developed for the culture of mice embryonic cells.Today, it finds a broad application of serum free culture of normal and transformed mouseand chicken cells. Ham’s Nutrient Mixtures were originally developed to support growth ofseveral clones of Chinese hamster ovary (CHO) cells, of HeLa and of mouse L-cells. Ham’smixtures were formulated for use with or without serum supplementation, depending onthe cell type being cultured. Ham’s F-12 was developed for growth of primary rathepatocytes and rat prostate epithelial cells. It is also used in a clonal toxicity assay usingCHO cells. The medium is for research use only and not approved for human or animaluse or for application in any in vitro diagnostic procedure. Storage of the medium andsupplements Please note that the basal medium has to be stored at 4°C up to 8°C butnever should be frozen. The supplement has to be stored at -20°C. Protect from light fora longer time period.Preparing and Stability:Thaw the supplement mixture at 37°C before adding to the basal medium. After thawingplease rinse the bottle and the tube with 70% ethanol as protection. Remove ethanolexcess before opening the cap of the tube. After adding the supplement mixture to thebasal medium slew carefully to get a homogenous medium. Please be careful not to touchthe tube or bottle interior threads with your fingers. Please avoid long term exposure tolight because some components are light sensitive. Also do not warm up the medium over37°C. All this protects your medium against contamination or degradation of essentialcomponents.
