General Information: Dulbecco’s MEM (DMEM) Ham’s F-12 w/o Arginine, Leucine, Lysineis the most widely used modification of BME. It contains a 4-fold higher concentration ofamino acids and vitamins. Non-essential amino acids and certain essential trace elementswere added. The bicarbonate concentration was increased. The standard formula forDMEM is with 1000mg/ml glucose. DMEM “High Glucose“ with 4500mg/ml aides in thecultivation of certain cell types. DMEM was originally developed for the culture of miceembryonic cells. Today, it finds a broad application of serum free culture of normal andtransformed mouse and chicken cells. Ham’s Nutrient Mixtures were originally developedto support growth of several clones of Chinese hamster ovary (CHO) cells, of HeLa and ofmouse L-cells. Ham’s mixtures were formulated for use with or without serumsupplementation, depending on the cell type being cultured. Ham’s F-12 was developed forgrowth of primary rat hepatocytes and rat prostate epithelial cells. It is also used in aclonal toxicity assay using CHO cells. The medium is for research use only and notapproved for human or animal use or for application in any in vitro diagnosticprocedure. Storage of the medium and supplements Please note that the basal mediumhas to be stored at 4°C up to 8°C but never should be frozen. The supplement has to bestored at -20°C. Protect from light for a longer time period.Preparing and Stability:Thaw the supplement mixture at 37°C before adding to the basal medium. After thawingplease rinse the bottle and the tube with 70% ethanol as protection. Remove ethanolexcess before opening the cap of the tube. After adding the supplement mixture to thebasal medium slew carefully to get a homogenous medium. Please be careful not to touchthe tube or bottle interior threads with your fingers. Please avoid long term exposure tolight because some components are light sensitive. Also do not warm up the medium over37°C. All this protects your medium against contamination or degradation of essentialcomponents.
